Nanoemulsion for Improving Antioxidant and Antibacterial Activity of Musa paradisiaca L. leaves Extract.

Abstract

Introduction: The ethyl acetate extract of Musa paradisiaca L. leaves contains quercetin. The main problem of quercetin associated with oral administration is its poor aqueous solubility. Nanoemulsion is drug delivery system that it was prove to enhancement the solubility and permeation of lipophilic drugs. The research aimed to investigate nanoemulsion as carrier of extract to improve the activity of antioxidant and antibacterial. Methods: The nanoemulsion of extract was formulated by various concentration of Tween 80 and PEG 400, and its was optimized by Factorial Design. The antioxidant and antibacterial activity of nanoemulsion and extract were determined by DPPH and Kirby Bauer methods, respectively, and its analyzed by independent T-test. Results: The optimum formula was obtained by 15% Tween-80 and 10% PEG-400. It has pH 6.4, droplet size 15.3 nm, a polydispersion index (PI) 0.012, a zeta potential of -23.3 mV, and stable during stability tests. The zone of inhibition of nanoemulsion is 14 mm againts S. Aureus and 10.33 mm againts E. Coli, and it exhibited a greater antibacterial acitivity than extract, having a zone 8.7 mm and 7.3 mm. Similarly, the nanoemulsion showed a better antioxidant activity at 29.52 ppm than extract at 54.69 ppm. The independent T test showed that nanoemulsion significantly improved antioxidant and antibacterial activities againts S. Aureus and E. Coli with p value 0.001, 0.003, and 0.000 respectively. The results prove that nanoemulsion succesfully improve antioxidant and antibacterial activity.
