Abstract
The study of drug interaction of glibenclamide and amla fruit extract (EBM) was carried out to obtain scientific data for considering the safety used of both. 2 research design is used to assess the pharmacodynamic and pharmacokinetic interactions of glibenclamide and EBM. First, 18 rats were divided into 6 groups, normal control, glibenclamide control, 2 control doses of EBM 500 and 750mg/kg, and 2 test groups given glibenclamide and EBM with 2 different doses of 500 and 750mg/kg. A glucose tolerance test is used to analyze the effects of glibenclamide. Second, 15 rats divided into 3 groups, glibenclamide control, and 2 test groups were given glibenclamide and EBM with 2 different doses of 500 and 750mg/kg. Glibenclamide levels are determined at the specified time intervals, measured using a spectrophotometer. The pharmacokinetic profile of glibenclamide was determined by linear pharmacokinetic analysis with the compartment method using PKSolver. Glibenclamide experienced drug interactions when used with EBM at a dose of 750mg/kg. The onset of drug interactions was observed at 90 minutes, hypoglycemia effect increased 7,5 times. The mechanism of drug interaction occurs in the pharmacokinetic process but not absorption, Cmax and AUC glibenclamide were observed to increase after being used with amla fruit extracts.
